Lactobacillus reuteri in healthy subjects increases the level of serum 25OH vitamin D that influences calcium absorption and benefits bone health. The mechanism through which this probiotic influences vitamin D level is not clear. This may be due to a modification in the gut environment that specifically favors vitamin D absorption or to indirect effect on increased hepatic 25-hydroxylase activity or 7-dehydrocholesterol concentration, due to reduced absorption of dietary and biliary cholesterol .

Another possible mechanism through which GM influences bone health is mediated by its effect on the production of gut serotonin . In the recent past a dual effect of serotonin in the regulation of bone mass has been described, depending on the site of production of this molecule . A role as a bone mass regulator was proposed to gut-derived 5HT , which is influenced by GM. Enterochromaffin cells of the duodenum are responsible for the synthesis of g5HT, which is partially modulated by GM, as SCFAs increase the synthesis of g5HT . It has been shown that 5HT interacts with bone cells and, in particular, decreases osteoblast proliferation. These observations suggest that regulation of g5HT by GM may be a potential therapeutic strategy to improve bone health.

We recently reported that L. reuteri counteracts age-associated sarcopenia- and cancer-induced cachexia . The beneficial effects on skeletal muscle coexisted with increased growth hormone levels, decreased systemic inflammatory tone, and increased Forkhead Box N1 expression in the thymus gland. FoxN1-deficient mice fail to inhibit sarcopenia after L. reuteri consumption, suggesting a FoxN1-mediated mechanism .

Oxytocin has also a trophic effect in skeletal muscle . Experiments in mice show that plasma levels of oxytocin naturally decrease with age and that the hormone is necessary for muscle tissue regeneration. Oxytocin-deficient mice develop premature sarcopenia, while the administration of an oxytocin antagonist reduces muscle regeneration. By contrast, systemic administration of oxytocin activates the MAPK/ERK signaling pathway to promote aged muscle stem cell activation/proliferation and muscle regeneration .

Several studies in mice demonstrate that L. reuteri increases bone density and prevents bone loss in both menopausal ovariectomized and type 1 diabetes mouse models . In the female-ovariectomized mice, the L. reuteri effects are mediated by the suppression of effector CD4 + T-cell-mediated osteoclastogenesis . In the male diabetic mice L. reuteri inhibited the TNF–induced reduction of osteoblast number and activity .

Growth Media And Culture Conditions

Both probiotic lactobacilli strains were routinely cultured on de Man, Rogosa and Sharpe agar and incubated anaerobically for 48 h at 37°C. Subsequently, lactobacilli strains were inoculated into MRS broth and incubated anaerobically for 24 h at 37°C in an orbital shaker . All of the C. glabrata strains were cultured on Yeast Extract-Peptone-Dextrose agar and incubated aerobically for 24 h at 37°C. Candida glabrata colonies were transferred into YPD broth and incubated aerobically for 24 h at 37°C in an orbital shaker .

Strains And Culture Conditions

The Candida albicans and bacterial strains used in this study are listed in Table 1. Stock cultures of all strains were stored at 80°C with 15% v/v glycerol as cryopreservative. C. albicans was routinely grown in YPD media . For coculture assays, C. albicans was grown in the same media as lactobacilli and staphylococci, that is, Difco Lactobacilli MRS broth . Regular agar plates contained 15g/L Agar while 7g/L of agar were used in soft agar overlays. Standard incubation conditions for pure yeast cultures were 30°C and ambient aeration. Bacteria were grown at 37°C under a 5% CO2 atmosphere in a CO2 incubator. Bacteria-Candida cocultures were incubated as indicated in a CO2 incubator or in ambient air at 37°C growth temperature.

In Vitro Growth Inhibition

The rich MRS medium was chosen for these experiments because it supported vigorous growth by the Lactobacillus, Staphylococcus aureus, and C. albicans strains. Agar overlays of C. albicans on MRS plates inoculated with the probiotic lactobacilli and control bacteria represented a relatively simple screening method for growth inhibitory activities of the bacteria towards this yeast. The deferred assay regimen with a 2-day preincubation of the bacteria was the method of choice because simultaneous inoculation often led to no or very small bacterial colonies due to rapid overgrowth by the fungal cells.

Inhibition of C. albicans growth by lactic acid at low pH. Microplate growth assays were used to determine the effect of lactic acid on the growth of C. albicans. MRS medium was supplemented with 120mM lactic acid and the pH was adjusted to pH 4.5 or 7.0. MRS with 120mM lactic acid had a pH of 4.2. As a control for low pH, MRS was adjusted to pH 4.5 using hydrochloric acid ). While acidic pH slows growth of C. albicans, the strongest growth inhibition was achieved with lactic acid at low pH. This inhibitory effect was completely lost by neutralization. Error bars show the standard deviations of two experiments with quadruplicate measurements. Statistical significant differences to all other conditions as calculated by ANOVA are indicated by asterisks.

What Is Lactobacillus Rhamnosus

Lactobacillus rhamnosus is a bacterial species that belongs to genus Lactobacillus. Earlier it was considered to be a subspecies of Lactobacillus casei but recent genetic research concluded that it is a separate species in the L. casei clade. L. rhamnosus is a gram-positive, homofermentative, facultative anaerobic, rod-shaped, and non-spore-forming bacterial species. It often appears in a chain. Some strains of this species are used as probiotics. They are also useful in treating infections of the female urogenital tract such as bacterial vaginosis.

They are helpful to control dysbiotic overgrowth of other bacteria during the infection period. This bacterial species is sometimes used in dairy products such as fermented milk and long ripened cheese. Though this species is considered to be beneficial, it may not be beneficial to some subsets of the population. This is because in rare circumstances, it causes endocarditis in those who have weakened immunity and infants. The L Reuteri Probiotics I Recommend

Not all Lactobacillus reuteri probiotics are created equal. There are multiple different strains of Lactobacillus reuteri, which have different effects.

The precise strain that raised testosterone in the MIT rat study was Lactobacillus reuteri ATCC 6475. Other reuteri strains may or may not have the same effect it is impossible to say for sure before further studies are carried out.

ATCC 6475 is not the most common strain in probiotic supplements, but . A Swedish probiotic manufacturer called BioGaia actually owns the patent for ATCC 6475.

While I havent yet tested BioGaias probiotics myself, I plan to do so in the near future. I have taken another L. reuteri probiotic called Primadophilus on several occasions. It contains another well documented L. reuteri strain, and also carries Lactobacillus rhamnosus which had the positive effect on cortisol levels.

Probiotics are regarded as generally safe for most people. Nevertheless, there is always a risk of adverse effects with any supplement or dietary change. People who suffer from histamine intolerance or immunosuppression may have an increased risk of harmful side effects from probiotic intake.

Beneficial bacteria are also found in certain foods yogurt, sauerkraut, kombucha, kimchi and aged cheese all contain a high number of probiotic microbes.
