Growth Media And Culture Conditions

Both probiotic lactobacilli strains were routinely cultured on de Man, Rogosa and Sharpe agar and incubated anaerobically for 48 h at 37°C. Subsequently, lactobacilli strains were inoculated into MRS broth and incubated anaerobically for 24 h at 37°C in an orbital shaker . All of the C. glabrata strains were cultured on Yeast Extract-Peptone-Dextrose agar and incubated aerobically for 24 h at 37°C. At enrolment and each follow-up time point, two vaginal samples were collected by specimen collection swabs Technology Co., Ltd., Guangdong, China): one for the screening of BV by Nugent scoring and to measure vaginal pH, and the other was stored in 3 ml of buffer Technology Co., Ltd, Guangdong, China) at -80°C and later used for vaginal microbiota DNA profiling. All the patients were asked to self-collect a sample of faeces at enrolment and each follow-up visit using a self-collection bag containing the transport medium from BGI. The faecal samples were also stored at -80°C immediately upon receipt at the laboratory.

Total DNA of vaginal and faecal samples was extracted using a QIAamp DNA Mini Kit and DNA concertation was determined by a Qubit Fluorometer, and the purity and integrity of the gDNA were measured by 0.6% agarose gel electrophoresis. The DNA samples were stored at -80°C for further usage. Qualified genomic DNA samples were configured with the specific primers 8F and 518R , and the variable regions V1 to V3 of the 16S rRNA gene were amplified by PCR. Subsequently, Agencourt AMPure XP magnetic beads were used to purify PCR amplicons, which were dissolved in TE buffer and labelled to complete library construction. An Agilent 2100 Bioanalyzer was used to detect the insert fragment size, and then the products were sequenced on the HiSeq2500 platform using the PE300 module.

Growth Inhibitory Activity Of Probiotic Lactobacilli Strains Against Candida Glabrata

The growth inhibitory activity of probiotic lactobacilli strains was demonstrated as both Lact. rhamnosus GR-1 and Lact. reuteri RC-14 strains produced visible inhibition zones against all of the tested C. glabrata strains. In addition, C. glabrata ATCC 2001 was the most sensitive strain to the growth inhibitory effects of Lact. rhamnosus GR-1 and Lact. reuteri RC-14, with the largest inhibition zones of 1·63 ± 0·04 and 1·54 ± 0·04, respectively .1). To investigate whether pH contributes to the growth inhibitory effects of probiotic lactobacilli strains, the pH of MRS agar was buffered and neutralized by the addition of MOPS to pH 7·0. The probiotic Lact. rhamnosus GR-1 and Lact. reuteri RC-14 strains still managed to produce visible inhibition zones against all of the tested C. glabrata strains despite the buffering of the growth media with MOPS to a pH of 7·0 before inoculation. However, the visible inhibition zones appeared to be smaller than those of the previous experiments that utilized unbuffered MRS agar .1). The growth inhibitory activity of Lact. rhamnosus GR-1 and Lact. reuteri RC-14 against the tested C. glabrata strains was reduced by 2356% and 2062%, respectively, when the MRS media were neutralized to pH 7·0.

Bacterial Strains And Growth Conditions

Probiotique Lactobacillus Rhamnosus

L. rhamnosus GR-1 and GG were cultured on de Man Rogosa Sharp agar anaerobically using anaerobic packs at 37°C for 24 h under static condition. For cell culture challenge, lactobacilli were grown from a 1% inoculum in MRS broth for 24 h followed by washing and resuspending in the original volume with phosphate buffered saline . Uropathogenic E. coli GR12 was grown in Luria-Bertani medium at 37°C and constant shaking. Heat-killed bacteria were prepared by washing cultures in PBS and heating at 70°C for 1 h followed by plating 100 l on the respective growth medium to confirm loss of viability. Heat-killed L. rhamnosus GR-1 and E. coli were stored at -20°C until used for cell challenge.

Study Groups And Randomization

A total of 328 women between 12- and 16 weeks of gestation consented and were screened between May and October for the presence of an intermediate or high Nugent score . Eighty-six women whose vaginal samples had a Nugent score ⥠4 were randomized.

A Z test was used to determine the sample size with alpha = 0.05 and power = 0.8, giving a per-group sample size of 40 women to detect a difference between an asymptomatic Intermediate/BV prevalence of 30% in the probiotic group and 60% in the placebo group at the end of treatment protocol. This was increased to 43 in each group to compensate for 5% lost to follow-up. Subjects were randomized using a web-based service to receive two identical looking capsules per day containing either GR-1 and RC-14 or placebo given orally for 12 weeks. Vaginal swabs were collected at 13-, 28- and 35 weeks of gestation and analyzed for Nugent score, cytokines, chemokines and microbiota. Fourteen subjects were lost to follow-up or withdrew, 3 had taken less than 25% of the capsules, and there was insufficient sample for analysis in 3 others. This left 32 subjects in the probiotic and 34 in the placebo group with samples available for sequencing analysis and samples from 31 probiotic and 33 placebo subjects for cytokines and chemokine analysis.

Dna Isolation And Polymerase Chain Reaction Amplification Of V6 Region Of 16s Rdna

Vaginal swabs were equilibrated in 800μL phosphate buffer saline on ice and vortexed for 1 min. DNA was extracted with a Qiagen Stool Extraction Kit , bacterial DNA was amplified with barcoded primers targeting the V6 region of the 16S rDNA and PCR amplification was performed with colorless GO-Taq hot start master mix for 25 repeating cycles of 95 °C, 55 °C and 72 °C for 1 minute each step. The amplified products were quantified using a QuBit broad-range double-stranded DNA fluorometric quantitation reagent kit . Samples were pooled at equal molar concentrations and purified using Wizard PCR Clean-Up Kits .
