Role In Maintaining Vaginal Health

Since a vaginal microbiota is dominated by Lactobacillus species in the majority of healthy women, the idea of administering Lactobacillus probiotics to restore and/or maintain health was perpetuated. The intent was not to colonize the niche but to use strains that would restore the health status of the area. Various studies have been performed to investigate the role of LGR-1 for the treatment and prevention of vaginal disorders (which has had almost no new therapeutic approaches in over 40

Vaginal And Faecal Microbiota Sample Processing And Analysis

Probiotics for Women’s Health – Chapter 10

At enrolment and each follow-up time point, two vaginal samples were collected by specimen collection swabs Technology Co., Ltd., Guangdong, China): one for the screening of BV by Nugent scoring and to measure vaginal pH, and the other was stored in 3 ml of buffer Technology Co., Ltd, Guangdong, China) at -80°C and later used for vaginal microbiota DNA profiling. All the patients were asked to self-collect a sample of faeces at enrolment and each follow-up visit using a self-collection bag containing the transport medium from BGI. The faecal samples were also stored at -80°C immediately upon receipt at the laboratory.

Total DNA of vaginal and faecal samples was extracted using a QIAamp DNA Mini Kit and DNA concertation was determined by a Qubit Fluorometer, and the purity and integrity of the gDNA were measured by 0.6% agarose gel electrophoresis. The DNA samples were stored at -80°C for further usage. Qualified genomic DNA samples were configured with the specific primers 8F and 518R , and the variable regions V1 to V3 of the 16S rRNA gene were amplified by PCR. Subsequently, Agencourt AMPure XP magnetic beads were used to purify PCR amplicons, which were dissolved in TE buffer and labelled to complete library construction. An Agilent 2100 Bioanalyzer was used to detect the insert fragment size, and then the products were sequenced on the HiSeq2500 platform using the PE300 module.

What Does Research On L Rhamnosus Gr

Some studies show that it is very helpful for a vaginal infection such as a vaginal yeast infection or bacterial vaginosis, and also for a urinary tract infection .

For more information on the specifics of the probiotic Lactobacillus rhamnosus GR-1, including details of what studies have shown for the strain and at which dosages and duration, look for a future publication, “The Lactobacillus rhamnosus GR-1 Guide.”

Can Lactobacillus Rhamnosus Gr

Lactobacillus rhamnosus GR-1 is a lactic acid bacteria from the genus Lactobacillus and the species rhamnosus. Like other Lactobacillus rhamnosus, it is a Gram-positive, rod-shaped bacterium which does not produce spores and is not capable of moving itself with flagella.

L. rhamnosus GR-1 was isolated from the urethra of a healthy woman. The urethra is a tube that connects the urinary bladder to the urinary meatus for the removal of fluids from a woman’s body. Dr. Gregor Reid and Dr. Andrew Bruce at Urex Biotech discovered and developed Lactobacillus rhamnosus, GR-1® and then sold the rights to Chr. Hansen of Denmark.

What Are The Benefits Of Lactobacillus Rhamnosus

Lactobacillus Rhamnosus pack of 60 capsules * Epigenetics

Lactobacillus rhamnosus has been the focus of many research projects with some strains showing to be particularly beneficial for supporting female intimate health conditions. These certain strains of L. rhamnosus support vaginal health in the following ways:

  • Colonising in the vaginal tract Colonising primarily in the vaginal tract rather than the intestines, therefore supporting vaginal health .
  • Attachment to the vaginal mucosa By adhering to the urethra, certain strains of L. rhamnosus will displace and prevent the adhesion of pathogens to the vaginal lining which protects against infections such as bacterial vaginosis .

You can read more about this in our article on our sister site, Probiotics Learning Lab: Probiotics and bacterial vaginosis

Lactobacillus rhamnosus also supports digestive and immune health in the following ways:

  • Production of antimicrobial substances This helps to alleviate diarrhoea that can be associated with travel or antibiotic use .
  • Reduction of inflammatory signalling in the intestines – Reducing the frequency of IBS and IBD related abdominal pain. .
  • Modulation of the immune system By increasing the action of T-lymphocytes and the action of anti-inflammatory cytokines. .

Is L Rhamnosus Gr

The Lactobacillus rhamnosus species as a whole are listed on the European QPS list. Lactobacillus bacteria as a whole are considered to be routinely encountered in the environment, in fermented foods and drinks, and are frequently isolated from feces. Unless a person is immune-compromised, Lactobacillus are generally considered safe. If you ever have any questions about your immune status, please talk with your doctor.

Medical Research And Use

While L. rhamnosus GG is able to survive the acid and bile of the stomach and intestine, is claimed to colonize the digestive tract, and to balance intestinal microbiota, evidence suggests that L. rhamnosus, comparable to virtually all probiotic lactobacilli, is only a transient inhabitant and not autochthonous. Regardless, it is considered a probiotic useful for the treatment of various maladies, as it works on many levels. Lactobacillus rhamnosus GG binds to the gut mucosa.

Lacticaseibacillus rhamnosus GG is beneficial in the prevention of rotavirusdiarrhea in children. Prevention and treatment of various types of diarrhea have been shown in children and in adults.L. rhamnosus GG can be beneficial in the prevention of antibiotic-associated diarrhea and nosocomial diarrhea and this has been recently supported by European guidelines.Lactobacillus rhamnosus GG may reduce the risk of traveler’s diarrhea.

Acute gastroenteritis

A position paper published by ESPGHAN Working Group for Probiotics and Prebiotics based on a systematic review and randomized controlled trials suggested that L. rhamnosus GG may be considered in the management of children with acute gastroenteritis in addition to rehydration therapy.

Growth Inhibitory Activity Of Probiotic Lactobacilli Strains Against Candida Glabrata

The growth inhibitory activity of probiotic lactobacilli strains was demonstrated as both Lact. rhamnosus GR-1 and Lact. reuteri RC-14 strains produced visible inhibition zones against all of the tested C. glabrata strains. In addition, C. glabrata ATCC 2001 was the most sensitive strain to the growth inhibitory effects of Lact. rhamnosus GR-1 and Lact. reuteri RC-14, with the largest inhibition zones of 1·63 ± 0·04 and 1·54 ± 0·04, respectively .1). To investigate whether pH contributes to the growth inhibitory effects of probiotic lactobacilli strains, the pH of MRS agar was buffered and neutralized by the addition of MOPS to pH 7·0. The probiotic Lact. rhamnosus GR-1 and Lact. reuteri RC-14 strains still managed to produce visible inhibition zones against all of the tested C. glabrata strains despite the buffering of the growth media with MOPS to a pH of 7·0 before inoculation. However, the visible inhibition zones appeared to be smaller than those of the previous experiments that utilized unbuffered MRS agar .1). The growth inhibitory activity of Lact. rhamnosus GR-1 and Lact. reuteri RC-14 against the tested C. glabrata strains was reduced by 2356% and 2062%, respectively, when the MRS media were neutralized to pH 7·0.

Study Groups And Randomization

A total of 328 women between 12- and 16 weeks of gestation consented and were screened between May and October for the presence of an intermediate or high Nugent score . Eighty-six women whose vaginal samples had a Nugent score ⥠4 were randomized.

Consort flow chart of pregnant women enrolled in the study.

A Z test was used to determine the sample size with alpha = 0.05 and power = 0.8, giving a per-group sample size of 40 women to detect a difference between an asymptomatic Intermediate/BV prevalence of 30% in the probiotic group and 60% in the placebo group at the end of treatment protocol. This was increased to 43 in each group to compensate for 5% lost to follow-up. Subjects were randomized using a web-based service to receive two identical looking capsules per day containing either GR-1 and RC-14 or placebo given orally for 12 weeks. Vaginal swabs were collected at 13-, 28- and 35 weeks of gestation and analyzed for Nugent score, cytokines, chemokines and microbiota. Fourteen subjects were lost to follow-up or withdrew, 3 had taken less than 25% of the capsules, and there was insufficient sample for analysis in 3 others. This left 32 subjects in the probiotic and 34 in the placebo group with samples available for sequencing analysis and samples from 31 probiotic and 33 placebo subjects for cytokines and chemokine analysis.
